# What is DSIP studied for?

DSIP, delta sleep-inducing peptide, is a nine-amino-acid peptide first isolated from rabbit blood and later studied in people for sleep and insomnia. As of 23 September 2026, the human studies are small and mixed: a controlled insomnia study found weak effects, and no gene or receptor for DSIP has been identified.

## What it is

Delta sleep-inducing peptide is a nine-amino-acid peptide first isolated from rabbit blood in 1977. No gene, precursor protein or receptor for it has been identified, so its mechanism of action is unknown, and a 2006 review describes its link to sleep as poorly characterised.

## Human studies

In a 1992 placebo-controlled study of 16 people with chronic insomnia, sleep efficiency was higher and sleep onset shorter than on placebo, but the authors judged the effects weak and not of major therapeutic benefit.

In a 1981 placebo-controlled crossover study of 6 healthy volunteers, total sleep time in the following 130 minutes rose by a median 59% compared with placebo.

In a randomised crossover study of 11 healthy men, DSIP lowered blood ACTH-like activity for at least 3 hours with no change in cortisol; a 1995 placebo-controlled study found no effect on ACTH or cortisol.

## Side effects reported

In a small randomised study during anaesthesia, DSIP was associated with a higher heart rate and lighter anaesthetic depth than saline; the size of the change is not given in the abstract.

An uncontrolled 1984 series of withdrawal patients reported headache in a few patients, without a rate.

Neurosci Behav Physiol 1998: In rats with restricted blood flow to the brain, DSIP reduced posture problems and deaths.

Seizure 2005: In rats, DSIP and an analogue reduced the incidence and duration of induced seizures.
