# MOTS-c Explained: Mitochondrial Peptide Research (Philippines)

Your mitochondria have their own DNA. In 2015, scientists found it hides a tiny signaling peptide called MOTS-c. MOTS-c is a mitochondrial-derived peptide, just 16 amino acids long, encoded inside a mitochondrial gene called 12S rRNA. It was described in Cell Metabolism in 2015.

In cell studies, MOTS-c slows the folate cycle. That activates AMPK, the cell's energy sensor. Its main target appears to be skeletal muscle. In that 2015 study, mice given MOTS-c did not develop the insulin resistance seen with aging or a high-fat diet, and gained less weight on that diet.

These were mouse results. A 2021 study in Nature Communications linked it to exercise. In humans, exercise raised MOTS-c in muscle about 12-fold. In old mice, MOTS-c treatment roughly doubled running time.

The closest human treatment data comes from an analog, not MOTS-c itself. In a 2021 phase 1b trial, 20 adults with obesity and fatty liver took it for four weeks. Blood glucose fell modestly versus placebo. Liver fat fell similarly in both groups.

What is unknown? Almost everything in people: long-term safety, real-world effects, and whether mouse results translate. Status: MOTS-c is not an approved medicine anywhere. Research-grade vials are for research only.
