# NAD+ Explained: NR, NMN, IV and What Trials Show (Philippines)

NAD+ is a coenzyme, not a peptide. Here is what human trials on NAD+, NR and NMN actually found. Research notes

First, a correction. NAD+ is on almost every peptide menu, but it is not a peptide at all. NAD+ stands for nicotinamide adenine dinucleotide. It is a coenzyme, two nucleotides joined together, found in every living cell.

It is not built from amino acids. Its job: carrying electrons in energy metabolism, and fuelling enzymes such as sirtuins and PARPs that help regulate and repair cells. Why the interest? A 2012 study of human skin samples found NAD+ levels fell with age in both men and women.

Most human trials test precursors, NR and NMN, that the body converts into NAD+. A 2018 crossover trial in 30 healthy middle-aged and older adults found NR raised NAD+ in blood cells by about 60%, and it was well tolerated. But results on health outcomes are modest. A 2018 trial in 40 obese men found NR did not improve insulin sensitivity.

A 2021 trial in 25 prediabetic women found NMN improved muscle insulin sensitivity. Direct IV NAD+ is popular at wellness clinics, but evidence is thin. A small 2026 review of six clients reported cramping, nausea and chest pressure during infusions. NAD+ is not an FDA-approved drug.
