# PT-141 (Bremelanotide) Explained: The Clinical Trials

Most peptides in this space never became medicines. PT-141 did. So what did the trials actually show? PT-141 is the code name for bremelanotide, a synthetic peptide.

It is an active metabolite of an older compound, melanotan II. Bremelanotide is a melanocortin receptor agonist, acting mainly on the MC3 and MC4 receptors. The FDA notes that exactly how this affects desire is unknown. The evidence comes from two phase 3 trials: 24 weeks, randomised and placebo controlled.

The trials included 1,247 premenopausal women with acquired, generalized hypoactive sexual desire disorder. About 25% on bremelanotide had a meaningful rise in desire scores, against 17% on placebo. There was no difference in the count of satisfying events. Nausea was the most common side effect, reported by 40% of participants.

Flushing, injection site reactions, headache and vomiting also occurred. Blood pressure rose briefly after dosing. About 1% saw darkening of the skin or gums. In June 2019, the US FDA approved bremelanotide as a prescription medicine for this condition in premenopausal women only.
