The unit scale on an insulin syringe is just a volume scale wearing a different name. Once that clicks, every conversion in peptide preparation becomes one multiplication.

What a unit actually is

U-100 printed on a syringe means the scale divides each milliliter into 100 units. One unit is 0.01ml, ten units are 0.1ml, and a full 1ml barrel is 100 units. The naming comes from insulin, which is standardized at 100 IU per milliliter, but nothing about the syringe knows or cares what liquid it holds. For laboratory peptide preparation, the unit scale is simply the finest volume graduation cheaply available, which is exactly why these syringes are the standard tool for the job.

From concentration to micrograms per unit

The syringe measures volume. What that volume contains depends on how the vial was reconstituted. The conversion is one step: micrograms per unit equals the concentration in mg/ml times ten. A 10mg vial with 2ml of bacteriostatic water is 5 mg/ml, so one unit carries 50 mcg. Read the last column carefully. At 20 mg/ml, a 250 mcg draw sits at 1.25 units, a mark that does not exist on most barrels. At 2.5 mg/ml the same draw sits at a clean 10 units. This is the whole argument for reconstituting with more volume when target quantities are small: the graduation gets finer relative to the draw. Choosing a syringe size

Insulin syringes come in 0.3ml, 0.5ml and 1ml barrels, which read 30, 50 and 100 units. The physical length of the scale is similar across all three, so the smaller barrels stretch fewer units across the same distance: a 0.3ml syringe puts visible half-unit marks where a 1ml syringe crowds five units into the same space.
