TRIUMPH-2
In TRIUMPH-2, a phase 3 trial of retatrutide in adults with type 2 diabetes and obesity or overweight (n=1,152), mean body weight fell 12.7% on 4 mg, 19.1% on 9 mg and 20.8% on 12 mg over 80 weeks, against 4.0% on placebo (efficacy estimand; Eli Lilly topline release, 23 July 2026).

Results
A1C fell 1.4, 1.6 and 1.5 points from a baseline of 7.7% on 4, 9 and 12 mg, against 0.2 on placebo.
Side effects reported
In TRIUMPH-2, discontinuation because of adverse events was 3.8% on 4 mg, 11.6% on 9 mg and 7.7% on 12 mg, against 4.9% on placebo. The most frequently reported adverse events were diarrhoea in 27.4% on 4 mg, 33.5% on 9 mg and 33.6% on 12 mg against 13.2% on placebo, nausea in 13.7% on 4 mg, 20.8% on 9 mg and 28.0% on 12 mg against 8.0% on placebo, constipation in 14.0% on 4 mg, 16.2% on 9 mg and 16.8% on 12 mg against 9.4% on placebo and decreased appetite in 5.8% on 4 mg, 12.3% on 9 mg and 17.1% on 12 mg against 4.5% on placebo.
